What happened?
A Nature Aging study analysed longitudinal plasma samples from adults who achieved roughly 14% calorie restriction over two years in the CALERIE trial. The investigators linked calorie restriction to changes in the circulating exoproteome and found complement-pathway deactivation associated with lower inflammaging.
Why does it matter?
Chronic low-grade inflammation is one proposed contributor to age-related decline. Human mechanistic data are valuable because many longevity claims are based only on animals or cell models.
How to interpret the evidence
This is mechanistic evidence, not proof that calorie restriction extends human lifespan. Excessive restriction can be harmful, and the CALERIE intervention was controlled and moderate.
What remains uncertain?
It does not justify extreme fasting, malnutrition or claiming that calorie restriction is a proven human life-extension therapy.
