Oral aleniglipron reaches double-digit weight loss in phase 2b study

What happened?

The ACCESS phase 2b trial randomized 230 adults with overweight or obesity to once-daily aleniglipron or placebo. At week 36, placebo-adjusted mean weight change ranged from −8.2% to −11.3% across active-dose groups, without an apparent weight-loss plateau at the end of the double-blind period.

Why does it matter?

Aleniglipron is another example of the shift toward oral small-molecule incretin drugs, which could change cost, supply and patient preference if later trials confirm benefit.

How to interpret the evidence

Treatment-related discontinuation occurred and gastrointestinal effects remained important. Larger and longer phase III trials are still required.

What remains uncertain?

It is not yet an approved alternative to semaglutide, tirzepatide or orforglipron.
